Phospholipase B Domain Containing 2 (PLBL2) is a protein-coding gene found on the human chromosome 12q24.13.

PLBL2 is also referred to as P76. A 65472 Da protein comprising 589 amino acids is encoded by the human PLBL2 gene. The PLBL2 protein is universally expressed in various tissues, including ovary and adrenal tissues.

While PLBL2 is associated with hydrolase activity, PLBD1 is a crucial paralog of the PLBL2 gene. PLBL2 is linked to certain diseases, such as Gallbladder Papillomatosis and Gm1-Gangliosidosis, Type Iii.

Background information of PLBL2/PLBD2

The PLBL2 gene localizes to the lysosome since its absence may lead to an unrecognized yet serious lysosomal storage disease. Both PLBD1 and PLBL2 are semi-orphans, which means they are likely to be the phospholipases of B class but with vague physiological substrates and thus functionalities.

The PLBD1 and PLBL2 genes represent a small gene family (sequence homology class) within the vertebrate species although one that manifests extended in some early diverging eukaryotes.

The PLBL2 gene poses a unique difficulty in that a series of post-translational steps are evidently required for its activation. Without these steps, promising substrates can seldom be tested.

Such post-translational steps involve mannosylation suitable for the lysosome targeting receptor, the removal of the signal peptide, and self-catalytic proteolytic activation to expose the binding site of the substrate as this turns out to be appropriate.
