Interleukin-21 (IL-21): Recombinant cytokines and receptors for further study

The cytokine IL-21 is primarily generated by CD4+ T cells and natural killer T (NKT) cells. T cell, B cell, natural killer (NK) cell, macrophage, and dendritic cell (DC) proliferation and differentiation need IL-21/IL-21 receptor (IL-21R) signaling.

The IL-21/IL-21R axis is important in viral infections, cancer, and a variety of immunological disorders, including inflammatory bowel disease (IBD), rheumatoid arthritis, and type 1 diabetes.

IL21 belongs to the γ chain cytokine family, which also includes IL2, IL4, IL7, IL9, and IL15. The JAK1 and JAK3 pathways are activated when IL21 binds to IL21R and the common chain (γc/CD132/IL2RG), which then activates the STATs (STAT1/STAT3/STAT5a/STAT5b), phosphoinositide 3-kinase (PI3K/Akt), and MAP kinase pathways.

IL2RA (CD25), IL2RB (CD122), IL4R (CD124), IL7R (CD127), and IL9R are all members of the γc family of cytokine receptors (CD129).
