Carbonic anhydrases: Isoforms as therapeutic targets for cancer

The bicarbonate and proton concentrations, which regulate the pH of the cell, are crucially regulated by carbonic anhydrases (CAs). This modulation is essential for cell survival and proliferation, In solid tumors, CA proteins IX and XII’s activity is inhibited by sulfonamide-based inhibitors and monoclonal antibodies.

These inhibitors and antibodies are performing well in clinical trials for cancer, including one that Memorial Sloan Kettering Cancer Center is launching this year that combines a CA IX inhibitor and Nivolumab.

There is also evidence of dysregulation of all other catalytically active isoforms of CA (I, II, III, IV, CAs VA-VB, CAs VI-VII, CAs XIII, and XIV) in various cancer types. Following further investigation, they might eventually be used as diagnostic and prognostic markers.
